内容記述 | Sea buckthorn is a deciduous shrub of the Elaeagnaceae family native to Europe and Asia. Sea buckthorn berry (SBB) is pale yellow to dark orange in color, rich in nutrients, and many other researchers have reported a variety of biological functions associated with them. In this study, we have investigated the suppressive effect of sea buckthorn berry extract and its fractions on Type-I allergy using mast cell lines in vitro. An SBB fraction containing the highest amount of antioxidant polyphenols significantly inhibited the release of chemical mediators, such as histamine and leukotriene (LT) B4, from stimulated mast cells. This fraction also inhibited Ca2+ influx and phosphorylation of tyrosine residues of proteins, including spleen tyrosine kinase (Syk), which is associated with the signal transduction of chemical mediators released from stimulated mast cells. And we identified isorhamnetin, the predominant flavonol aglycon in this fraction. Isorhamnetin inhibited the histamine and LTB4 release from stimulated cells and suppressed intracellular Ca2+ influx. These results suggest that isorhamnetin is the primary substance responsible for the anti-allergic activity in SBB. In conclusion, SBB may alleviate Type-I allergy by inhibiting the release of chemical mediators from mast cells, and the polyphenols may contribute to this effect. | |||||||
